The present invention is directed to a low cost shieldable safety needle assembly. The assembly includes a needle cannula and a tip guard axially movable along the needle cannula through a drive mechanism. The drive mechanism may be interconnected between the tip guard and the needle cannula through a hub. The tip guard is axially movable along the needle cannula from a proximal position substantially adjacent a proximal end of the needle cannula at the hub, to a distal position in which the tip guard protectively surrounds the distal end of the needle cannula, thus effectively shielding the puncture tip of the needle. The drive mechanism is a unitary structure which is capable of maintaining a first selfsupporting shape for maintaining the tip guard in the proximal position and is deflectable from the first self-supporting shape to a second extended shape in which the tip guard is moved to the distal position. The drive mechanism may be a rigid flexible planar sheet material which includes a plurality of folds for defining the selfsupporting shape. THE USE OF BEOWULF CLUSTER TO ACCELERATE THE PERFORMANCE OF MONTE-CARLO PARAMETRIC EXPECTATION MAXIMIZATION MCPEM ; ALGORITHM IN ANALYZING COMPLEX POPULATION PHARMACOKINETIC PHARMACODYNAMIC EFFICACY DATA. BACKGROUND: Development of population pharmacokinetic PK ; pharmacodynamic PD ; efficacy model used for the clinical trial simulation in drug development is resource and time intensive. OBJECTIVE: This study describes the use of Beowulf cluster to accelerate the performance of MCPEM algorithm in developing complex population PK PD efficacy model. METHODS: The Beowulf cluster consisted of six Pentium 4 computers connected with gigabit ethernet switch. A MC-PEM algorithm in S-ADAPT program was written in Fortran and run on Windows XP Professional operating system. A PK PD efficacy data of efalizumab from 240 psoriasis patients was used to develop the complex mechanism-based population PK PD efficacy model. The model consisted of six differential equations and 22 parameters and fit simultaneously to the data. The performance of the cluster was considered satisfactory and have a good scalability if the parallel efficiency time to complete a single iteration by a single node time to complete a single iteration by all six cluster nodes number of cluster nodes is 0.5. RESULTS: Depending on the loading size for each computer node ranged from 5-40 ; , the parallel efficiency ranged from 0.49 to 0.90. This corresponding to inefficiency of 0.51 to 0.10 that was mainly due to communication overhead and load imbalance between different cluster nodes. CONCLUSION: The Beowulf cluster is able to accelerate the performance of MCPEM algorithm in analyzing the complex population PK PD efficacy data.

Hallucinogens also known as psychedelics, act on the central nervous system to produce significant, often radical, changes to the user's state of consciousness; can distort the user's sense of reality, time and emotions. Nevertheless, most of our knowledge about the architecture of the skins nervous system is originating from the middle of the 20th century and still surprisingly little is known about its three-dimensional topography. We used dermal sheet preparations, immunohistochemical staining thereof with antibodies to PgP9.5 and NCAM CD56 and subsequent confocal laser scanning microscopy in order to analyse the intact nerve end organ in situ in three dimensions. Both antibodies showed a regular network of fibers throughout the entire superficial dermis. Staining for PgP9.5 was weaker and nerve fibers were not decorated continuously, but in a bead like fashion. NCAM CD56 staining additionally presented protrusions with diameters ranging from 5 to 15m. In further immuno-electron microscopic analysis, we detected evidence for ultrastructural features of Schwann cells ensheating terminal nerve endings. These endings appeared to end freely and spread out within 25m below the dermo-epidermal junction. To visualize the topographic relationship of the nerve end organ with other skin components, double staining with NCAM CD56 and anti-vWF was done. Larger trunks of the nerve end organ were found to intertwine with the superficial capillary plexus but no obvious relation of terminal nerve endings and blood vessels could be found. Our novel approach will help to further characterize the nerve end organ under physiologic conditions as well as during diseases of the skin. The fact, that nearly all penicillate nerve endings spread out below the dermo-epidermal junction should make us think again about the presence of nerve fibers in the epidermis.
